    DMSO 属于非质子极性溶剂，常用于化学反应、 PCR反应以及在细胞、组织和器官的保存中用作玻璃化低温冷冻防护剂。 DMSO用于细胞冷冻培养基内，可以保护细胞免受冰晶引起的机械性损伤。它也可以用于主培养、亚培养、重组异倍体、杂交瘤等系列细胞株，胚胎干细胞（ ESC）和造血干细胞的冷冻保藏。另外常常与 BSA或FBS混合使用。

用  途：

（1） DMSO广泛应用在人、动物细胞株及细菌噬菌体λ的低温防护中，制备 DMSO溶液用来冷冻细胞的步骤如下：

A.配制冷冻培养基：含有细胞培养用培养基、 10-20% 血清和 5-10% DMSO；

B.用胰酶或其他合适的方法消化贴壁细胞；（细胞zui好处于对数增长期）

C.低速离心（4℃，250×g, 10min）收集细胞，吸去培养基；

D.用冷冻培养基悬浮细胞，使细胞密度为 106 -107 cells/ml；

E.平均分配细胞，装入冻存管内；

F.依照标准冻存程序冷冻细胞，冻存在 -70℃或更低的温度内；

（2）细胞融合时，40-50% PEG 溶液内加入10% DMSO；

（3）有文献报道 DMSO作为缓冲液的成分，用于以下的实验：Poly（A+）RNA 的纯化， E. coli感受态细胞转化、 PCR反应、 cDNA文库的扩增、 DNA测序、 DEAE-葡聚糖介导的细胞转染、聚凝胺介导的 DNA转染；

（4）有相应的程序描述到用 DMSO回收膜过滤后的 DNA，用于接下来的 PCR扩增反应；

（5）用于 DNA测序的毛细管电泳技术将 5% DMSO (v/w)混入 2 M尿素中，如果需要可以调整 DMSO浓度到100%。

（6）有研究报道不同浓度的 DMSO与寡核苷酸的熔点温度关系；

（7）有研究描述利用 DMSO提高杂交瘤细胞中单克隆抗体的产量； 调查发现低温冷冻前用含有 DMSO的培养基高温培养杂交瘤细胞。

（8）文献报道利用 DMSO修饰蛋白中的磷酸丝氨酸和磷酸苏氨酸残基用作磷酸化状态的质谱分析（ MS）；

（9）文献报道水和 DMSO中的亮脯利特（Leuprolide）降解作用；

注意事项：

    仅用于实验室，不可用于药品、家居和其他用处 。

使用说明：

    DMSO 易于水混合（ 1 ml DMSO + 1 ml H2O）形成清澈、无色的溶液。易吸潮，免暴露在潮湿环境。

储存 /稳定性：

DMSO 容易过冷并且室温下缓慢融化，收到的产品应为固体并非液体状态。固化的 DMSO放在室温下可以再次液化，对其本身无损害。属于热稳定化合物，在高达 100℃的酸、碱及中性溶液中保持稳定。温度接近其沸点（189℃），在碱性和中性溶液中仍保持稳定。 150℃加热 24h，其纯度损失小于0.1%。

要制备滤膜除菌的 DMSO溶液，推荐使用 teflon或 nylon膜，尽量不用醋酸纤维素膜。
